    Well I had my first test on monday......chapters 1,2,4,5, & 7. Got a 96% A :hatparty: :hatparty: :hatparty: :hatparty: :hatparty: :hatparty: Anyway, all I studied were the lecture notes that the teacher handed to us in the beginning of the semester. She did tell us that all the questions on the test were going to come from those notes....boy was she telling the truth. This class is going to be a breeze!!!!!!

    What are you doing in lab? Will you do staining? Will you find an unknown? Or is it just looking at pictures and slides? Even our lab was very difficult the aspect of actually doing all those different procedures and tests made it quite interesting and we learned sooooooooo very much.

    For lecture today, we went over Glycolysis, Kreb's Cycle, and Electron Transport Chain, as well as Aerobic, Anareobic and Fermentation.

    For lab we did 6 Gram-Stains of 6 different bacteria from the 3 different cultures we cultivated. Over the last few labs, we had grown our very own bacteria, stained them and observed them over the microscope to differentiate between morphologies, arrangement, gram (+) and gram (-).

    LOL this class is fun!

    I made an 86, but I am kind of happy cause there was only one A, and he said the grades werent very good. I dont think I will complain.

    i am so frustrated.....yesterday in lab we were doing gram stains. for some reason or another i am having a difficult time with the gram negative stains, they are supposed to come out a red/pinkish color which some of mine do, but then there is some blue mixed into it.

    i spoke to my prof. and she insists that the cause is due to me not rinsing the crystal violet completely off. i do feel that i did rinse it efficiently; however, i am worried that the class prior to mine may not be following procedures resulting in them contaminating the gram negative bacteria with the gram positive bacteria.

    what should i do? i followed the staining procedures to a 't'......any advice would be greatly appreciated.

    I am loving Micro so far I love the labs they are so interesting and so layed back, I am having a great time this semester albeit I am dead busy!

    I am glad that you are enjoying Micro. I am also very busy. I am taking Ethics and Human Growth ( I love the class) Ethics is so so.....

    I got back my first test today. The class average was about a 62. I got a 78. Everyone was shocked because we all took the test and thought we did really well... well, by everyone I mean my group of friends. Nobody got in the 90s, at all. The highest was an 88, which was my best friend in the class.

    Oh well, time to start studying for the next test!

    Hope you all did better.

    (he doesn't grade on a curve )

    LOL. No, the only A was a 100, so no curve. Oh, well, I will study harder from here on out.

    a reason why some gram + (purple) may appear gram - (pink) may be due to heat-fixing the smear too long causing some of the cell walls in those bacteria to rupture.

    another could be that you didn't allow the smear you prepared to air-dry completely. then when you heat-fixed it, the water still there causes the gram (+) cell wall to rupture, therefore not holding the primary stain intact.

    another would be that you let the decolorizer sit for too long.

    on another note, making a smear too thick may cause a smear to stain both colors. this is because the decolorizer penetrates through some, but not all. leaving the decolorizer sit for too short may cause some gram (-) cells to apear gram (+).

    it could also be what you stated above; the other class not following proper protocol. those were just suggestions i wrote above, hopefully it helps. how were your other slides?
