I am in my second semester out of four. I have had a really great experience with the best clinical instructor this semester, and I don't think I will have a chance to work with her again before graduation. So, I am wondering if it is appropriate to ask her for a letter of recommendation this semester instead of before graduation so I am "fresh" in her mind?

Educators rarely forget students. However, I would at the very least tell her what that you enjoyed your experience with her and would like to keep her for a future reference. Because in two years a letter dated 2015 isn't as effective as one that has a current date on it. Also, who would she address the letter to? "To whom it may concern" isn't always the best idea

This is very true. I e-mailed a former instructor about the program he is teaching at (he switched schools) and he was happy to hear from me. I graduated in 2009.

Heck yeah. I asked after first semester all three professors for letters and they were more than happy to write one. You want to build that portfolio up. It will look good in your interview if you show letters of recommendation from each years professors as well. Get as many as you can. All of mine have said they would update then as needed also. Good professors always want to help good students. Don't be afraid to ask.
