My town does one every year - it's hosted by a town organization, not the school but it is based at the school. I haven't gotten any reports of anyone having an exacerbation of asthma or difficulty breathing d/t the powder -and trust me when I say that I would be the one to get the calls from parents - not the town hall. The streets look so pretty afterward until it rains.
I've done the Color Run twice when it came to town. It was a lot of fun. It was a company that came in and did it, hosted by someone and profits went to a group. I have asthma. The powder is not aimed at the face typically, but yes, its a cornstarch base and it flies, especially in our wind. I have more asthma problems because my fat self was jogging than because of the powder. It is set up in intervals, every so far you get a new color tossed at you, so you are very rainbow colored by the time you finish. Big party at the end. Lots of fun :) Bad asthmatics could wear a mask going through the colored powder if it is a problem, but I'd worry more about the pollens in the air, personally.

This.
We had our first one this year for 5th grade - part of a fundraiser thru PTA. I was nervous about it but it turned out ok - I did have a few "asthma" issues but I would have had them with a regular fun run also. Last year for Field day and our big annual fundraiser of Boosterthon I started to be more proactive - sending a letter home to parents about sunscreen, allergy & asthma medication and basically letting them know I do not have any of this in my clinic and that they need to plan ahead for possible issues.
