Not a great idea. THe VP probably won't even look at it. Her secretary will forward it to the Recruiter.
... and if she does look at it, it would need to be extraordinary in some way. Anything less would be perceived as an annoyance and might make you look foolish.
If you happen to know her ... or meet her in some other context ... it's VERY appropriate to tell her of your interest. But for people to start sending unsolicited resumes out of the blue rather than going through the proper channels, most busy VP's would just be annoyed.

Would it be appropriate/inappropriate/waste of time/can't hurt to send a resume and cover letter outlining my strong desire to join the organization as a staff RN?
This would be for a major hospital, not like a DON of a nursing home, in other words, an administrator that doesn't normally have a hand in hiring staff RN's.
