Disease we don't know cause and drugs we don't know how it works question

Hello, got two questions I need some help on.

• What are some of the diseases or illnesses that we do not know the cause of? Also is Alzhema one of them?
  
• What are some drugs where we know it works, but we don't know how/why it works? I mean, it just works. Please exculde psychiatric related drugs.

MS? They think it might be an autoimmune disorder. But last I checked nobody knows for sure what can trigger it.

Last I heard in pharmacology, nobody knows how aspirin works. They obviously know it does, but don't have any real idea of how. It just does. There were other drugs, but I can't remember off the top of my head.

I learned those a few years ago, so things might have changed since.
