I was wondering as a Toronto Nurse (rpn) if I would apply to the transition program in vancouver if they would accept me? I go on there website and saw you have to be a lpn in the province of BC. Does that mean I would have to do the rpn BC test, to then qualify or am I missing something.

There is no BC test for PNs. It's the same test for all the provinces except Quebec and you only have to pass it once. You would just need to get a BC LPN license.
How would you apply for one, Is there centers in Toronto where we can go write them? Also can I hold on to mutilple provinces lpn licenses, such as Ontario and Vancouver at the same time to increase my chances to get in a university?
Go to write what? You don't have to write anything. Do you have your RPN license in Ontario? Once you have that all you need to do for a BC license is apply, pay the fee and meet the provinces requirements. Multiple licenses will not affect getting into university.

Once you have met provincial requirements, passed the exam and have a license issued you can apply to another provincial college for a license. As mentioned the exam is accepted by all provinces except Quebec so you only have to sit it the once and pass
